The Introduction of Dutasteride

Dutasteride is a new dual inhibitor of 5α-reductase that inhibits both 5α-reductase-1 and 5α-reductase-2. It is more effective than finasteride in reducing DHT concentrations (94.7% vs. 70.8%). Dutasteride inhibited 5α-reductase-1 60 times more than finasteride. The incidence of prostate cancer was 50% lower in the dutasteride group than in the placebo group 27 months after dutasteride administration.

Dutasteride is a second-generation alpha 5 reductase inhibitor approved in the United States for the treatment of benign prostatic hyperplasia. Unlike alpha1-adrenoceptor antagonists, which work immediately, dutasteride takes several months to work on prostate size and symptoms of prostate enlargement (urinary hesitation and dyspareunia).

Dutasteride  inhibits the growth of the prostate in patients with benign prostatic hyperplasia (BPH) by inhibiting type I and II 5α-reductase isozymes. The effect of this product on reducing DHT peaks after 1 to 2 weeks of use at 0.5 mg per day, and plasma DHT can be reduced by 85% to 90%. In patients with an enlarged prostate gland, 5mg per day was used for 12 weeks and DHT was significantly reduced.

Dutasteride is rapidly absorbed orally, bioavailability of about 60%, oral 0.5mg, 2-3h peak blood concentration, with food, peak concentration (Cmax) reduced by 10%-15%. The performance distribution volume of this product is 300-500L, the plasma protein binding rate is up to 99%, and the α1-acidic glycoprotein binding rate is 96.6%. The drug concentration in semen of healthy volunteers was 3.4ng/ml, similar to plasma. The plasma drug concentration reached steady state after 6 months of continuous use of this product. It is extensively metabolized by hepatic cytochrome CYP3A4 and excreted mainly by feces, of which 5% is unmetabolized drug and 40% is metabolites, and less than 1% is unmetabolized drug in urine. The elimination half-life of this product is about 5 weeks .

The Application & Function of Dutasteride

Dutasteride may be used to treat moderate to severe symptoms of benign prostatic hyperplasia (BPH). It is used in patients with moderate to severe symptoms of BPH to reduce the risk of acute urinary retention (AUR) and surgery.
