The Introductction of Virginiamycin(PRISTINAMYCIN)

Virginiamycin, also called PRISTINAMYCIN, is a streptomycin antibiotic, produced by Streptomyces sp. of PRISTINAMYCINII (PII), with PRISTINAMYCINI being a cyclic hexapeptide lactone and PRISTINAMYCINII being a polyunsaturated cyclic lactone, with PIA and PIIA being the major components of PRISTINAMYCIN. The in vitro antibacterial activity of the combination of the two components is at least 10 times higher than that of the single component, and this synergistic effect also expands the antibacterial spectrum of the two components so that they exhibit bactericidal effects against bacteria, while each component alone exhibits bacteriostatic effects.

At the same time, due to the presence of two components with completely different structures, resistance is acquired more slowly, which is one of the remarkable features of this new drug that distinguishes it from other anti-drug-resistant bacteria drugs, and it is the only orally administered anti-drug-resistant bacteria drug. PRISTINAMYCIN has the appearance of a light yellow powder with a non-crystalline structure and a melting point of 151153°C. PRISTINAMYCIN has a cyclophore of [a]D25 = 130° in 0.5 mol/L methanol and [a]D25 = 150° in 1 mol/L trichloromethane.

PRISTINAMYCIN is soluble in methanol, ethanol, ethyl acetate and trichloromethane, slightly soluble in water and insoluble in petroleum . PRISTINAMYCIN is oxidized in Tollens reagent, Millon reagent, Seliwanoff reagent, Fehling reagent and in hydrated xunotrione, but reduced in ferric chloride solution, Ehrlich reagent and FolinDenis reagent. PRISTINAMYCIN has been used in France for more than 20 years as an anti-staphylococcal oral preparation, however, the staphylococci and enterococci resistant to PRISTINAMYCIN are less than 5%, and so far, no Gram-positive cocci have been reported to be resistant to puromycin.

PRISTINAMYCIN, as a vancomycin alternative, is currently one of the most effective antibiotics for the treatment of infections caused by resistant Gram-positive bacteria.PRISTINAMYCIN mainly acts on bacterial ribosomes and inhibits protein synthesis. However, its clinical application is limited by the fact that PRISTINAMYCIN is extremely poorly water soluble and cannot be easily made into parenteral formulations.

It can be used in pig feed at a dosage of 10 (10.2 million units) to 20 (20.4 million units) g/t; it can also be used in chicken feed below 16 weeks of age at a dosage of 2 to 5 g/t, and is prohibited during the egg-laying period. It has inhibitory effect on gram-positive bacteria and can prevent and control bacterial dysentery (such as pig dysentery, chicken necrotizing enteritis, etc.), with almost no drug resistance.
