The Introduction of Mesalamine
Masalazine, also known as 5-aminosalicylic acid, is an organic compound with the chemical formula C7H7NO3 and is the active ingredient in SASP for the treatment of ulcerative colitis. It has a significant inhibitory effect on inflammation of the intestinal wall. Mesalazine inhibits the synthesis of inflammation-causing prostaglandins and the formation of the inflammatory mediator leukotrienes, thus acting as a significant inhibitor of inflammation in the intestinal mucosa. Mesalazine inhibits prostaglandin synthesis in a dose-dependent manner and reduces the release of PGE2 in the human colon mucosa.
Masalazine is a white crystalline powder with a melting point of about 280°C (decomposition). It is soluble in hydrochloric acid, slightly soluble in hot water, slightly r soluble in cold water or ethanol.
The Application & function of Mesalamine
Due to the high levels of prostaglandin E2 (PGE2) and leukotriene B4 (LTB4) in the colonic mucosa of patients with active UC.
1. Masalazine inhibits prostaglandin synthesis and reduces PGE2 release in human colonic mucosa in a dose-dependent manner.
2. Masalazine also inhibits lipoxygenase activity in neutrophils. Addition of mesalamine to cultured human colonic mucosal cells resulted in dose-effect inhibition of two important inflammatory mediators, chemotactic LTB4 and leukotrienes. At high doses, mesalamine was able to inhibit certain functions of human neutrophils, such as migration, degranulation, phagocytosis and oxygen radical synthesis.
3. Masalazine also inhibits the synthesis of platelet activity factor (PAF), which plays an important role in the development of inflammation. It is more effective in the connective tissue of the intestinal wall with inflammation. It is used in ulcerative colitis, ulcerative proctitis and Crohn's disease.
4. It has a significant inhibitory effect on inflammation of the intestinal wall. Mesalazine inhibits the synthesis of prostaglandins that cause inflammation and the formation of leukotrienes, an inflammatory mediator, and thus has a significant inhibitory effect on the inflammation of the intestinal mucosa.
