The Introductction of Chloroquine Phosphate
Chloroquine phosphate is white crystalline powder, odorless, bitter taste. Gradually change color when exposed to light; aqueous solution shows acidic reaction. It is soluble in water, almost insoluble in ethanol, trichloromethane and ethe*. Melting point is 193~196℃, decomposed at the same time when melting.
Chloroquine phosphate belongs to 4-aminoquinoline antimalarials, which can inhibit the nucleic acid synthesis of Plasmodium falciparum schizonts, interfere with the growth and reproduction of Plasmodium falciparum, and have high efficient killing effect on Plasmodium falciparum intra-erythrocytic stage schizonts.
Chloroquine phosphate has a strong binding force with nuclear proteins, and through the proximity of the negatively charged 7-chloro group on its quinoline ring to the 2-amino group on DNA guanine, chloroquine is inserted between the two strands of the double helix of DNA. It forms a complex with DNA, thus preventing DNA replication and RNA transcription. Chloroquine also inhibits phosphoric acid doping of Plasmodium DNA and RNA, interfering with Plasmodium reproduction due to reduced nucleic acid synthesis.
The Application & Function of Chloroquine Phosphate
Chloroquine phosphate is used for the treatment of chloroquine-sensitive Plasmodium falciparum, Plasmodium vivax, and Plasmodium triple day malaria.
Chloroquine phosphate is used for suppressive prophylaxis of malaria symptoms.
Chloroquine phosphate is also used in the treatment of parenteral amebiasis, connective tissue diseases, and photosensitivity diseases (e.g., sun erythema).
