The Introductction of Mitomycin C

 

Mitomycin C is a broad-spectrum antitumor antibiotic extracted from Streptomyces bacterium (bacteria), which is genotoxic and antitumorigenic, and is a universally recognized inhibitor of DNA damage, with anticancer effects on a wide range of cancers, and upon activation, acts as a DNA alkylating agent, which acts on the principle of depolymerization of the cellular DNA, and selectively inhibits DNA, cellular RNA and protein synthesis, thus inhibiting tumor cell division.


Mitomycin C is a blue-purple lustrous crystal or crystalline powder. Solid stable, but easily inactivated in acidic and alkaline solution.Mp>360℃; the maximum absorption wavelength in methanol is 216nm, 360nm and 560nm, and the maximum absorption wavelength in aqueous solution is 365nm±2nm.This product is soluble in organic solvents, such as water, methanol, aceton* and ethyl acetate; it is slightly soluble in benzene, ethyl ethe* and carbon tetrachloride, and insoluble in petroleum ethe*. Highly toxic, LD50 (rat, oral) 14mg/kg.


Mitomycin C has a wide anti-tumor spectrum and is effective in gastric cancer and breast cancer. It is effective in lung cancer, liver cancer, malignant lymphoma, Hodgkin's disease, reticulocyte sarcoma, uterine cancer, leukemia, intestinal cancer and pancreatic cancer, but the remission period is shorter. Combined with urokinase can improve the efficacy, the product acts rapidly, but the number of therapeutic operators is not high and the toxicity is high. The product and bleomycin and its derivative pailomycin are DNA-damaging antibiotic anticancer drugs, which can depolymerize cellular DNA and inhibit the replication of DNA in proliferating cells. The intravenous LD50 in mice is 5 ml/kg.

Mitomycin C is a cell cycle non-specific drug, which has wider anti-tumor spectrum and rapid action, but with low therapeutic index and higher toxicity. It is clinically applicable to digestive tract cancers, such as gastric cancer, intestinal cancer, hepatocellular carcinoma and pancreatic cancer, with good efficacy. It is also effective in lung cancer, breast cancer, cervical cancer and chorionic epithelial cancer. It can also be used for malignant lymphoma, cancerous pleural and abdominal effusion. Mitomycin C is also a chemical mutagen that can cause DNA mutation. Saffron extract can significantly inhibit the reproductive toxicity produced by anticancer drugs such as cisplatin, cyclophosphamide, mitomycin C and urethane, while the magnitude of inhibitory effect is not significantly related to the dose. It is foreseeable that the cooperative use of saffron with other anticancer drugs is expected to reduce the toxic side effects of these anticancer drugs.
