The Introductction of Clotrimazole
Clotrimazole is a synthetic pyrrole antifungal drug, which has inhibitory effect on a variety of pathogenic fungi, and can also have fungicidal effect at high concentration. Clotrimazole inhibits the biosynthesis of sterols such as fungal ergosterol by interfering with the activity of cytochrome P450; it damages fungal cell membranes and changes their permeability, causing leakage of important intracellular substances; it also inhibits the biosynthesis of fungal triglycerides and phospholipids; it also inhibits the activity of oxidation and peroxidase, causing excessive accumulation of peroxides in cells, resulting in structural degeneration and necrosis of fungal subcellular structures. Clotrimazole has broad-spectrum antifungal activity, and has good antibacterial effect on S. epidermidis, S. trichiura, S. trichiura, S. stainum, Cryptococcus spp. and Candida spp. It also has certain antibacterial activity on S. schenckii, S. dermatitidis, S. vulgaris, S. histolytica, etc.
Clotrimazole is white powder or colorless crystalline powder. Melting point 147-149℃. Soluble in anhydrous ethanol, aceton*, chloroform, almost insoluble in water. Odorless, tasteless, decomposes rapidly in acid solution. Clotrimazole hydrochloride, melting point 159℃.
In vivo and in vitro clotrimazole has good antibacterial effect on Candida, Cryptococcus, Aspergillus, Alcaligenes and dermatophytes, the efficacy on superficial skin fungal disease is similar to ashwagandha, the efficacy on deep fungal disease is similar to amphotericin B. It has good effect on visceral pathogenic fungi such as Candida albicans, Cryptococcus novelis, Coccidioides and Histoplasma, etc. The effect on Candida albicans is stronger than mycoplasma It is stronger than mycoplasma. The fungus is not easy to produce resistance to this product.
Clotrimazole is easily absorbed internally and can be used internally to treat systemic and deep fungal infections, such as Aspergillosis, Candida albicans, Cryptococcosis, Coccidioidomycosis, Histoplasmosis (dogs, cats) and fungal sepsis. For severe deep fungal infections, it should be combined with amphotericin B. It can also be used externally to treat superficial fungal infections such as ringworm and ringworm of the body and hair of domestic animals. It is not effective for ringworm of the head. Clotrimazole is also effective against certain gram-positive bacteria and Trichomonas vaginalis.
The Application & Function of Clotrimazole
Clinical Applications
Clinical topical application of clotrimazole cream or solution 1% for the treatment of Candida ringworm, lichen planus, tinea pedis, ringworm of fingernails and other skin infections. Clotrimazole vaginal tablets (100mg) are administered to treat vaginal infections of Candida and Trichomonas. Side effects include local irritation and burning sensation.
Oral administration treats deep mycobacterial infections, but the toxic side effects are more severe, including gastrointestinal irritation, neutropenia and abnormal liver function.
1. Oral administration: (1) For the treatment of Candida infection in the oropharynx. 2) For the prevention of Candida infection in the oropharynx in patients with chemotherapy, immunocompromised or deficient.
2. Topical topical application: (1) Candida skin infections. (2) Tinea capitis, ringworm and ringworm of the body due to Trichophyton rubrum, Microsporum sp. and Tinea epidermidis; Tinea pedis due to Sporothrix furfur, etc.
