The Introductction of Chloramphenicol
Chloramphenicol is an antibiotic with the chemical formula of C11H12Cl2N2O5. It is easily soluble in methanol, ethanol, propanol and ethyl acetate, slightly soluble in ether and chloroform, and insoluble in petroleum ether and benzene. Chloramphenicol is extremely stable, and its aqueous solution does not fail after boiling for 5h. Because there are two asymmetric carbon atoms in chloramphenicol molecule, there are four optical isomers of chloramphenicol, of which only the left isomer has antibacterial ability
The Application & Function of Chloramphenicol
Chloramphenicol antibiotics can act on the 50S subunit of bacterial ribonucleosomes and block the synthesis of proteins. They are broad-spectrum antibiotics with antibacterial activity.
The 70S ribosomes of bacterial cells are the main cellular components of synthetic proteins, which include two subunits, 50S and 30S. Chloramphenicol blocks the action of transpeptidylase by reversibly binding to 50S subunit, interfering with the binding of aminoacyl-tRNA terminal with amino acid to 50S subunit, thus blocking the formation of new peptide chain and inhibiting protein synthesis. Because chloramphenicol can also bind to 70S of human mitochondria, it can also inhibit the protein synthesis of human mitochondria and cause toxicity to human body. Because the binding of chloramphenicol to 70S ribosomes is reversible, it is considered to be a bacteriostatic antibiotic, but it can also produce bactericidal effect on some bacteria at high drug concentration, and can produce bactericidal effect on influenza bacilli even at low concentration.
