The Introductction of Rapamycin
Rapamycin (RAPA) is a new type of macrolide immunosuppressant, a white solid crystal, melting point 183-185℃, lipophilic, soluble in methanol, ethanol, acetone, chloroform and other organic solvents, very slightly soluble in water, almost insoluble in ether.
Rapamycin (also known as "sirolimus") is a secondary metabolite secreted by Streptomyces soil from the soil of Easter Island, Chile for the first time in 1975, and its chemical structure belongs to the "triene macrolide" compounds.
Rapamycin is a macrolide antibiotic, similar in structure to plucofosfamide (FK506), but with very different immunosuppressive mechanisms. fK506 inhibits the proliferation of T lymphocytes from G0 to G1 phase, while RAPA blocks the progression of T lymphocytes and other cells from G1 to S phase by blocking signaling through different cytokine receptors, compared to FK506. RAPA blocks calcium-dependent and non-calcium-dependent signaling pathways in T lymphocytes and B lymphocytes.
The Application & Function of Rapamycin
Rapamycin, a targeted inhibitor of mTOR, is able to treat tumors closely related to this pathway including kidney, lymphoma, lung, liver, breast, neuroendocrine and gastric cancers. It is particularly effective in the treatment of LAM (lymphangioleiomyomatosis) and TSC (tuberous sclerosis), two rare diseases.
Rapamycin is a new immunosuppressive agent with good efficacy, low toxicity and no nephrotoxicity.
Rapamycin is now often used as a drug to maintain immunity in transplanted organs (especially kidney transplants) to slow down immune rejection after transplantation, however, scientists have recently discovered that it has.
Another use for rapamycin: it can be used to treat Alzheimer's disease (senile dementia).
