The Introductction of Metformin Hydrochloride
Metformin hydrochloride is an oral hypoglycemic agent of the biguanide class, also known as mefloquine hydrochloride, gevalt and metformin, which has a weaker hypoglycemic effect than phenelzine, and its main effect is to reduce the secretion of A-cell glucagon and promote the conversion of glucose into glycogen, with the property of not stimulating the secretion of insulin by pancreatic beta-cells.
Metformin hydrochloride is crystalline or crystalline powder. Melting point 232℃ (218-220℃). Soluble in water, dissolved in methanol, slightly soluble in ethanol, insoluble in ether and chloroform. Odorless, bitter taste.
The mechanism of action of metformin hydrochloride in lowering glucose is as follows:
1. Increase the sensitivity of peripheral tissues to insulin and increase insulin-mediated glucose utilization.
2. Increase glucose utilization in non-insulin-dependent tissues, such as brain, blood cells, renal medulla, intestine, skin, etc.
3. Inhibit hepatic glycogen allogenesis and reduce hepatic glucose output.
4. Inhibit glucose uptake by intestinal wall cells.
5. Inhibit cholesterol biosynthesis and storage, reduce blood triglyceride and total cholesterol levels.
The Application & Function of Metformin Hydrochloride
Clinically, metformin hydrochloride is mainly used to treat type II diabetic patients with unsatisfactory dietary control alone, especially those who are obese and with hyperinsulinemia, with this drug not only has a hypoglycemic effect, but also may have the effect of reducing body weight and hyperinsulinemia. It can be effective in some patients with poor efficacy of sulfonylurea, such as combining with sulfonylurea, small intestine glucosidase inhibitor or thiazolidinedione hypoglycemic agents, which is more effective than using them separately. It can also be used in patients on insulin therapy to reduce insulin dosage.
Adverse reactions
The incidence of adverse reactions of metformin hydrochloride is higher than that of methylsulfonylurea. Common gastrointestinal reactions include anorexia, nausea, vomiting, abdominal pain, diarrhea, and occasionally gastrointestinal reactions such as metallic taste in the mouth, bad breath, nausea, loss of appetite and diarrhea, and sometimes hypoglycemic symptoms are seen.
