The Introductction of GUANIDINE THIOCYANATE
Guanidine thiocyanate is white crystalline powder, melting point 116-121℃, PH value of 6mol/L in water at 20℃ is 4.5~6, solubility is completely dissolved.
Guanidine thiocyanate is an uncoupling agent, a very strong class of protein denaturant, which can dissolve proteins and make protein secondary structure disappear, cell structure degradation, and nucleoprotein rapidly separated from nucleic acid.
Guanidine thiocyanate is used for denaturing lysis of cells; extraction of rna and dna, without rnaase and dnaase activity. Guanidine thiocyanate and b-mercaptoethanol work together to inhibit rnase activity, while guanidine thiocyanate and sodium dodecyl sarcosinate work together to denature proteins, thus releasing rna; dna rarely dissociates under acidic conditions and is precipitated along with protein denaturation, while rna dissolves in the supernatant.
